{"id":966,"date":"2022-09-15T10:30:00","date_gmt":"2022-09-15T14:30:00","guid":{"rendered":"https:\/\/medicamentquebec.ca\/?p=966"},"modified":"2025-12-11T14:55:45","modified_gmt":"2025-12-11T19:55:45","slug":"medicament-quebec-soutient-the-rna-zipcode-platform-an-ai-guided-platform-for-precision-delivery-of-rna-based-therapeutics","status":"publish","type":"post","link":"https:\/\/medicamentquebec.ca\/index.php\/2022\/09\/15\/medicament-quebec-soutient-the-rna-zipcode-platform-an-ai-guided-platform-for-precision-delivery-of-rna-based-therapeutics\/","title":{"rendered":"M\u00e9dicament Qu\u00e9bec soutient \u00abThe RNA Zipcode Platform: An AI-Guided Platform for Precision Delivery of RNA-Based Therapeutics\u00bb"},"content":{"rendered":"\r\n<p><strong>Activit\u00e9s financ\u00e9es &#8211; 15 septembre 2022<\/strong><\/p>\r\n<p><!--more--><\/p>\r\n\r\n\r\n\r\n<p><strong>Dans le cadre de sa deuxi\u00e8me phase<\/strong>, M\u00e9dicament Qu\u00e9bec annonce l&#8217;octroi d&#8217;un financement de 1\u00a0500\u00a0000 $ pour l\u2019activit\u00e9 \u00ab\u00a0<strong><strong>The RNA Zipcode Platform: An AI-Guided Platform for Precision Delivery of RNA-Based Therapeutics<\/strong>\u00a0\u00bb<\/strong> qui sera dirig\u00e9e par <a href=\"https:\/\/www.ircm.qc.ca\/fr\/chercheurs\/eric-lecuyer\" target=\"_blank\" rel=\"noreferrer noopener\">Dr Eric L\u00e9cuyer<\/a>, Professeur de l&#8217;Institut de recherches cliniques de Montr\u00e9al (IRCM).<\/p>\r\n\r\n\r\n\r\n<p><strong>En quoi consiste l&#8217;activit\u00e9?<\/strong><\/p>\r\n\r\n\r\n\r\n<p>Le but de ce projet est d&#8217;amener \u00e0 maturit\u00e9 la \u00ab\u00a0Plateforme de Design de Zipcodes ARN\u00a0\u00bb prototype pour le d\u00e9veloppement d&#8217;outils de ciblage th\u00e9rapeutique ARN de nouvelle g\u00e9n\u00e9ration et d&#8217;exploiter le potentiel commercial de cette plateforme via la cr\u00e9ation d&#8217;un RNA Zipcode entreprise en d\u00e9marrage. La localisation, la stabilit\u00e9 et la traduction intracellulaires des mol\u00e9cules d&#8217;ARN cellulaires sont contr\u00f4l\u00e9es de mani\u00e8re exquise par des modules de r\u00e9gulation discrets cod\u00e9s dans la s\u00e9quence d&#8217;ARN, d\u00e9finis par leur s\u00e9quence et leurs caract\u00e9ristiques structurelles, que nous d\u00e9signons par \u00abZipcodes ARN\u00bb. En plus de dicter la fonction et le destin des ARN cellulaires qui les contiennent, il existe \u00e9galement un \u00e9norme potentiel pour que des variantes naturelles ou synth\u00e9tiques de ces \u00e9l\u00e9ments servent d&#8217;amplificateurs de mol\u00e9cules d&#8217;ARN th\u00e9rapeutiques afin de contr\u00f4ler avec pr\u00e9cision leur administration et leur fonction sous-cellulaires. Alors que les codes postaux d&#8217;ARN ont longtemps \u00e9t\u00e9 difficiles \u00e0 identifier, un pipeline puissant qui combine des tests de reportage Zipcode-Mapping (Zip-Map) massivement parall\u00e8les \u00e0 la pointe de la technologie avec des logiciels de bio-informatique et d&#8217;apprentissage automatique (AA) de pointe a \u00e9t\u00e9 d\u00e9velopp\u00e9 afin de cartographier les positions des codes postaux d&#8217;ARN dans des s\u00e9quences naturelles et concevoir des variantes synth\u00e9tiques capables de contr\u00f4ler la localisation, la stabilit\u00e9 et les propri\u00e9t\u00e9s de traduction de l&#8217;ARNm. Cette plate-forme a \u00e9t\u00e9 exploit\u00e9e pour mener l&#8217;enqu\u00eate la plus approfondie \u00e0 ce jour sur l&#8217;espace des s\u00e9quences naturelles sur des dizaines de milliers de g\u00e8nes (par exemple, 5&#8217;UTR, 3&#8217;UTR, lncRNA), d\u00e9finissant des milliers de codes postaux capables de moduler le comportement d&#8217;un ARNm cargo attach\u00e9. Gr\u00e2ce \u00e0 des algorithmes AA propri\u00e9taires, des codes postaux synth\u00e9tiques avec une activit\u00e9 am\u00e9lior\u00e9e par rapport aux homologues de s\u00e9quence naturelle ont \u00e9t\u00e9 con\u00e7us et des tests d\u00e9velopp\u00e9s pour tester la fonctionnalit\u00e9 des codes postaux en conjonction avec des cargaisons th\u00e9rapeutiques. Cette proposition permettra de d\u00e9montrer le potentiel d&#8217;am\u00e9lioration des codes postaux ARN en combinaison avec divers types de mol\u00e9cules d\u2019ARN th\u00e9rapeutiques, poursuivant les objectifs suivants :<\/p>\r\n\r\n\r\n\r\n<ol class=\"wp-block-list\" type=\"1\">\r\n<li>Optimiser des approches AA permettant le design de Zipcodes ARN synth\u00e9tiques modulant la localisation, la stabilit\u00e9 et la traduction d\u2019ARNm cargo;<\/li>\r\n<li>Valider la capacit\u00e9 de Zipcodes ARN \u00e0 moduler diff\u00e9rentes fonctionnalit\u00e9s d\u2019ARNm cargo par des essais <em>in vitro;<\/em><\/li>\r\n<li>\u00c9valuation de l\u2019activit\u00e9 de Zipcodes ARN gr\u00e2ce \u00e0 la cr\u00e9ation d\u2019une plateforme de validation d\u2019ARN th\u00e9rapeutiques <em>in vivo.<\/em><\/li>\r\n<\/ol>\r\n\r\n\r\n\r\n<p>Dans l&#8217;ensemble, l&#8217;initiative multidisciplinaire et interinstitutionnelle, impliquant des \u00e9quipes de recherche de l\u2019IRCM\/Universit\u00e9 de Montr\u00e9al et de l\u2019Universit\u00e9 McGill, permettra d\u2019am\u00e9liorer l&#8217;efficacit\u00e9 des th\u00e9rapies \u00e0 base d&#8217;ARN, offrant un niveau sup\u00e9rieur de sp\u00e9cificit\u00e9 en m\u00e9decine de pr\u00e9cision. De plus, il contribuera \u00e0 nucl\u00e9er les forces du Qu\u00e9bec dans la recherche et la th\u00e9rapie de l&#8217;ARN, permettant au Qu\u00e9bec de s&#8217;engager activement dans la r\u00e9volution de l&#8217;ARN. En stimulant l&#8217;innovation en mati\u00e8re d&#8217;ARN, la plateforme ouvrira la voie \u00e0 la structuration d&#8217;un nouveau pilier pour la bio\u00e9conomie
